I’m sorry that I don’t have extra time to elaborate, but I think you may have received a micro USB to USB adapter in the kit of accessories (button caps, drill guide, etc). You could use that to hook up your USB stick.
The wpa_supplicant.conf file won’t be there. I think that whenever you create it, it gets moved elsewhere on the next boot. You have to create it and then boot the machine.
With the wifikeyfile.txt option, you create it, boot up, use the RetroPie/WiFi menu, and then tell it to grab the info from that file.
